I tried to connect to my Mac Pro with Remoter (iPad) the other day only to discover that it wouldn't connect. it took a few reads of this site and discussion.apple.com but I eventually was able to get it to work over 3G.
then today, I needed to connect to my Mac Pro from the city and couldn't. perplexing. I came home and discovered the problem.
as the others above have said, if I put the screen to 'sleep' using a Hot Corner, Remoter is unable to connect to the Mac. if I then wake the screen with a mouse click or keyboard press, Remoter can connect.
if however I set Preferences/Energy Saver/Display Sleep to 1min and allow the screen to sleep 'on its own', then Remoter can connect to the Mac.
physically turning off the (LCD) screen does not affect Remoter's ability to connect to the Mac.
so... as long as I don't use the "Put Display to Sleep" Hot Corner, Remoter can connect to the Mac... almost...
if the screen is locked using the Security/Require password preference, and the Login window is displayed when Remoter makes contact with the Mac, the connection times out and the connection is lost.
